Worshipping in spirit (note not the Spirit) means from our very depth of being and the truth is not being hypocritical. The Holy Spirit will be there when we worship that way. The woman was arguing about institutions i.e. where to worship, but Jesus was talking about how to worship. Since the Cross, the Temple or any other geographical point has become irrelevant.
Then the passage you quote shows us that the woman was truly waiting for Messiah/Christ even though she was a sinful Samaritan. She was more perceptive than the Jewish leaders.
